Without details, we really can't help.

Logs, network traces and general work to isolate the cause. 

For example, does the same happen if you use Samba as a file server
only.  Samba 4.0 as a DC use the same file server (by default) as our
member server and standalone file server modes.  If you are able to
eliminate the DC component, and provide those details then it may be
easier to get the attention of a number of developers who focus
exclusively on that area.
