Check MTU sizes and what is the firewall doing with DF bit's? 
I had a bad upstream router, that dropped my DF bits to no DF and that
cause lots or re-transmissing on my RDP links, just trowing in few ideas..

Only on 1 DC ? 
samba-tool dbcheck --reindex   ;-) might help also, 
if you havent done that already.
